Myeloperoxidase (MPO) Peroxidation Activity Assay Kit is a rapid, simple, sensitive, and reliable colorimetric assay suitable for use as a high throughput MPO activity assay. In the MPO assay protocol, myeloperoxidase produces HClO from H2O2 and Cl–. Myeloperoxidase (MPO) is a peroxidase enzyme (EC 126.96.36.199) most abundantly present in neutrophil granulocytes. It is a green hemoprotein found in neutrophils and monocytes that catalyzes the reaction of hydrogen peroxide and halide ions to form cytotoxic acids and other intermediates that play a role in the oxygen-dependent killing of tumor cells and microorganisms. Its heme pigment causes the green color in secretions rich in neutrophils, such as some forms of mucus. Furthermore, it can oxidize tyrosine to a tyrosyl radical using hydrogen peroxide as an oxidizing agent. Thus measurement of MPO peroxidation activity is crucial for disease prediction and therapeutics. Celltechgen’s Myeloperoxidase (MPO) Peroxidation Activity Assay Kit, oxidizes a substrate to generate fluorescence (Ex/Em = 571/585 nm), which is directly proportional to total peroxidase activity in the sample. A specific MPO inhibitor is provided, which suppresses peroxidase activity due to MPO. This permits differentiation of MPO-mediated peroxidation from the activity of other peroxidases which may be present in the samples. Our kit provides a sensitive, quick and easy method for detecting MPO activity. The assay is high-throughput adaptable and can detect less than 2 µU of MPO activity. Store kit at -20°C protected from light. Allow Assay Buffer to warm to room temperature
