Streptavidin is a tetrameric biotin-binding protein purified from Streptomyces avidinii with a near neutral isoelectric point. Streptavidin is composed of 4 identical polypeptide chain subunits, which binds 4 moles of biotin per mole of protein with a high affinity (Kd ~10-15). Biotinylated antibodies, proteins, peptides, nucleic acids and other molecules or interaction complexes can be captured, immunoprecipitated, removed or purified from samples using this streptavidin resin.
Streptavidin-Cy3 can be used to detect the biotinylated antibodies in immunohistochemistry and immunocytochemistry. Cy3 is excited maximally at 554 nm, emits at 570 nm, and Cy3 has a molar extinction coefficient of 1.3 × 105 M. The average molecular weight of Streptavidin-Cy3 is 60 kD and Streptavidin alone is 52 kD.
Supplied: Phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide.
Usage: We recommend that the reagent should be titrated for optimal performance for each application. The concentration provided is based upon molecular mass of streptavidin independent of any additional molecular mass that might be added by the Cy3 conjugation.
Storage: The Streptavidin-Cy3 solution should be stored undiluted between 2°C and 8°C, and protected from prolonged exposure to light. Do not freeze.
